Nana started his career as commercial artist and visualizer. During his earlier period, he was discriminated by his looks. But with his true talents, he came to the mainstream. He have played the roles of hero, supporting artist, villain, as well as comedian. After his debut film Gaman, he did many Marathi films. He played the role of Nathuram Godse in the British television series Lord Mountbatten.
His first National Award as Best Supporting Actor was for his role in Parinda. Later he won the National award for Best Actor was for Krantiveer. He also won National Award for Best Villian. He voice acted for the character Shere Khan for the Hindi version of The Jungle Book. He also did playback singing for his movies Yeshwant, Wajood and Aanch.

The actress Tanushree Dutta filed a complaint against Patekar on sexual harassment. Her claim was that, it happened at the set of Horn Ok Please. She filed the case before several authorities. Her statement brought the Me Too movement to Bollywood.
